Screening for male-derived biological material from collected samples plays an important role in investigations of offenders, especially those related to sexual offenses.
The authors have developed a LAMP method that targets repetitive sequences of the Y chromosome to detect male DNA.
After extracting male DNA in about 10 minutes using “Kaneka Easy DNA Extraction Kit version 2 “, the We successfully amplified from 0.5 ng of male DNA at 61-67°C.
Under conditions of 61-67°C, male DNA was observed to be amplified in less than 10 min, and amplification was completed in 20 min.
In contrast, it was not detected in samples containing up to 10 ng of female DNA under the same conditions.
As little as 1 pg of male DNA can be detected in this LAMP reaction.There was a correlation between the amount of DNA and reaction time in the range of 1 pg to 10 ng of template DNA.
Furthermore, in order to apply the LAMP method to on-site screening tests for male DNA, the authors have developed a protocol using dyes that can detect the LAMP reaction in colorimetric fashion.
Using the protocol developed by the authors, detection from DNA extraction of samples (male blood and saliva) to the end of the process in about 30 minutes and does not require a lot of work or special equipment.
This allows for quick and easy screening of male-derived samples in forensic investigations.
