Cannabis sativa (C. sativa), a cannabis plant, is grown around the world for a variety of purposes and However, the cultivation and possession of these crops is regulated by law in many countries. Accurate testing methods are needed to crack down on illegal possession and cultivation.
As we previously reported on the identification of C. sativa using the isothermal nucleic acid amplification (LAMP) method, the The LAMP method can be used for on-site detection of C. sativa.
While the previously introduced protocol took approximately 90 minutes from sampling to detection, this study describes a protocol for on-site testing (this protocol) that can be completed in 30 minutes.

Extract the DNA of cannabis grass in about 10 minutes by using “Kaneka Easy DNA Extraction Kit version 2” , and then, in this protocol, C. sativa was determined by LAMP reaction under optimal conditions (63°C) within 20 minutes.
This protocol allows for the detection of cannabis DNA from 10 pg of purified DNA, with C. sativa (20 strains) and other samples (50 strains) were evaluated to determine the specificity of C. sativa.
This protocol, using “Kaneka Easy DNA Extraction Kit version 2” and “MyAbscope” , allows you to detect and extract DNA in just 30 minutes. C. sativa can be identified from various plant parts such as leaves, stems, roots, seeds and resins derived from C. sativa extracts.

By using “MyAbscope” , a portable device that can complete the process from DNA extraction to detection in a single unit, this protocol can be used for in-site detection. This method is expected to contribute to the regulation of C. sativa.

In this paper, the authors used “Kaneka Easy DNA Extraction Kit version 2” for DNA extraction of cannabis grass and also used “MyAbscope” is used to detect C. sativa by the LAMP reaction.
